A possible distant rellie on Ancestry says she has DNA matches with male people (direct or collateral I suppose) going back to the 1600s. And has a DNA match to someone born in the mid 1600s. Is this possible for recombinant DNA technology to be able to do this now?

That would probably be the only way lol. I think the matches on Ancestry are to be taken with a pinch of salt, especially with autosomal DNA as far back as the 1600s.
